(Abstract No. 397) Interpretive Summary: Technical Abstract: We evaluated the effects of gonadal steroids and uterine stromal fibroblast (US) coculture on RBP secretion by UE cells. Cells were isolated from the uteri of 6 pigs and seeded into Matrigel coated Millicell HA filters (UE) or 12 well culture plates (US). Polarized UE and US cells were then maintained in DMEM:F12 + 5% charcoal stripped Fetal Bovine Serum (FBS) for 48 h before half the UE cells were cocultured with US. Cells were then either maintained in DMEM:F12 + 5% FBS or in media containing estradiol 17B (E; 10-9M), progesterone (P; 10-7M) or E+P. Total media in the apical compartment and half the media in the basal compartment was changed daily for 5 days. Media was analyzed using a radioimmunoassay specific for porcine RBP. Polarized UE cells cocultured with US secreted more RBP (0.74+/-0.09 ng/ml) than UE cells cultured alone (0.54+/-0.09 ng/ml), however the difference was not significant (P>.1). Approximately 64% of total measurable RBP was localized to the apical compartment (apical=0.78+/-0.07 ng/ml; basal=0.44+/-0.07 ng/ml; P<.03). P (0.53+/-0.1 ng/ml) did not influence RBP secretion when compared to control cultures (0.63+/-0.1 ng/ml). Treatment of UE cells with E (0.84+/-0.1 ng/ml) increased (P<.01) RBP secretion, an effect that was not seen in the presence of both E and P (0.48+/- 0.1 ng/ml). Therefore, RBP is secreted mainly into the apical compartment and E stimulates RBP secretion by UE cells. (USDA 97388144157 to GRN) |
