Technical Abstract: The Hessian fly is a major insect pest of wheat and occurs in all wheat- growing areas of the United States and most other production areas of the world. The primary method of control is through genetically resistant wheat. However, the development of biotypes of the insect capable of surviving on formally resistant wheat presents a threat to the permanence of resistance. While Hessian fly populations have been sampled to monitor biotype changes, no molecular analyses of geographic populations have been conducted. We investigated regions of the mitochondrial genome of Hessian fly for variation using PCR-RFLP and direct DNA sequencing. Intraspecific variation was detected and differences in haplotype frequencies among geographic populations were compared. Results are related to diversity within geographic populations and biotype evolution.
