Interpretive Summary: A precise, reproducible method for determining the amount of amylose present in starch was needed. Older laboratory methods are generally colorimetric and give either an overestimation or underestimation of amylose because of interference by lipid material in the sample. The described method is not affected by lipids. Another important factor in favor of this method is safety. Older methods use carcinogenic chemicals (dimethylsulphoxide, DMSO) in the solubilization stage of sample preparation whereas, the described method uses Potassium Hydroxide (KOH), urea and deionized water to solubilize the starch and as eluant.

Technical Abstract: A high performance size exclusion chromatography (HPSEC) method has been developed and tested on numerous starch samples for the separation and determination of percent amylose and amylopectin. The method described uses KOH, urea, and 90 min heating at 100 deg C to completely sulubilize the starch. HPSEC is accomplished using deionized, distilled water as eluant and separation of amylose and amylopectin is achieved within 30 min using one column.
