Interpretive Summary:

Technical Abstract: Perilipins, a family of lipid droplet associated phosphoproteins which are highly phosphory by protein kinase A (PKA), are suggested to play a role in lipolysis. The predominant forms of perilipin in adipocytes are perilipin (peri)A, 57 kDa, and periB, 46 kDa, with periA being more abundant (50 fold greater) than periB. Previously we noticed a trend towards a decrease in PKA-stimulated lipolysis with overexpression of perilipin (Souza et al, JBC, 273, 1998). In these studies, we further investigated the effects of overexpression of perilipins on isoproterenol- stimulated lipolysis. Perilipins subsequently treated with isoproterenol to stimulate lipolysis. Western blot analysis and measurement of glycerol release demonstrated that 20-30 fold increase in periB expression resulted in a 50% reduction in isoproterenol-stimulated lipolysis and very little, if any, change in endogenous periB expression (n=4). We are presently investigating the mechanistic basis for these observations associated with perilipin overexpression. In summary, the data provides novel information about the relative roles of both perilipin B and perilipin A in PKA- stimulated lipolysis.
