Green pepper and zucchini squash are sensitive to chilling temperatures. However, the severity of chilling injury in these commodities was reduced by methyl jasmonate and superatmospheric oxygen. Low temperature storage also induced alternative oxidase (AOX) gene expression in cyanide-resistant respiration in green pepper. Complementary DNA fragment of AOX was isolated from green pepper and zucchini squash using degenerate PCR. Southern analysis will be shown to estimate the gene copy number in the genome. Northern analysis will be performed to determine the steady-state AOX transcript level. The response of AOX to methyl jasmonate and superatmospheric levels of oxygen in the chilling-sensitive green pepper and zucchini squash will be determined. |
