Interpretive Summary: Avian leukosis subgroup J virus (ALV-J) is an economically important virus that can cause cancer-like disease and other production problems in meat-type chickens. In an attempt to develop specific diagnostic tests for ALV-J, we used standard methods to obtain sufficient amount of ALV-J envelope glycoprotein, a specific component of the virus. Abundant expression of this glycoprotein was noted in specific cell cultures (insect cells) commonly used in the laboratory for such expression. This method of expressing this ALV-J component will enable scientists to develop specific diagnostic tests for ALV-J and therefore enhance the ability to design effective control programs.

Technical Abstract: Expression effect of envelope gene of avian leukosis virus (ALV-J) in Sf9 cells infected with recombinant baculovirus rBac-env was analyzed by immunofluorescent assay and immunoprecipitation. The results showed that recombinant envelope gene product was a glycosylated protein in tunicumycin treatment test. The production of recombinant envelope gene product was highly expressed in Sf9 cells infected with 800 viruses per cell. Some of recombinant product was secreted on the cells' surface, but most of them was fusion protein in the cells. It will be a good resource of envelope protein for diagnosis and control of ALV-J.
