Interpretive Summary: Technical Abstract: Introduction: Conjugated linoleic acid (CLA) improves body composition and alters adipose tissue in vivo and adipocytes in vitro through unknown mechanisms. These mechanisms may include circulatory factors (i.e., fatty acids, hormones). Objective: The aim of this study was to determine if serum from CLA fed pigs affects adipogenesis in vitro. Methods: Sows were fed a corn/soybean meal based diet with 0.83% soy oil (n=8) or a 0.5% CLA diet supplemented with 0.83% CLA-60, Conlinco (n=6) beginning on day 40 of gestation. Within 12-20 hours of birth, serum was collected from one female and one male pig per litter. Primary stromal-vascular cells of subcutaneous adipose tissue from 75 day old fetal pigs were cultured (n=3) and plated on 35mm culture dishes. Cells were maintained in DMEM + 5% pig serum for 6 days. Immunocytochemistry was performed and total cell number, lipid containing cell number, preadipocyte number, and lipid filling was determined per 10x microscopic field. Statistical differences were determined using ANOVA. Results: In 75 day fetal cultures, serum from CLA pigs increased total cell number (CLA,1040.96 ±67.2; CON, 884.38 ± 61.34 cells/dish; P <= 0.05) and suppressed relative preadipocyte number (CLA, 1.34 ± 0.24; CON, 2.14 ± 0.22; P <=0.05). No significant differences in lipid filling were observed. |
