Interpretive Summary:

Technical Abstract: The isolation of Salmonella and serovar identification were compared in 7 feedyard (FY) and 3 control playas of the Southern High Plains in the winter and summer. The objective was to isolate and identify all Salmonella serovars in the 10 playas. Water and muck samples were collected in duplicate from the North, South, East, and West sides of each playa. One-hundred ml of playa water were mixed with 100ml of a double strength enrichment media (Sulfa Brilliant Green broth + novobiocin 25 g/ml) and shaker incubated at 37 C for 24 hrs. Next, 100 ml of the incubated broth was processed as before for another 24 hrs at 37 C. The 2nd incubated broth was plated on 3 Gram negative selective agar media (MacConkey, Brilliant Green, and XLD) and incubated at 37 C for 48 hrs, Salmonella like colonies were inoculated into slant agars (LIA, XLD, TSI) and incubated at 37 C for 24 hrs. Salmonella suspect colonies were placed on blood agar for confirmation and serovar identification. No Salmonella isolates were recovered from the 3 control playas. Seven Salmonella enteria serovars were isolated from 5 of 7 playas in the summer and 13 Salmonella enteria serovars were isolated from 7 FY playas in the winter. A total of 296 isolation attempts (47 were positive for Salmonella) were made in the summer. A total of 288 isolation attempts (171 were positive for Salmonella) were made in the winter.
