Interpretive Summary: Technical Abstract: Commercial bacterins for the control of fowl cholera are comprised of P. multocida serotypes A:1, A:3, and A:4, which are the serotypes that are most often associated with outbreaks in poultry flocks. A membrane-associated protein fraction (cross-protective factor, CPF), comprised of 11 major proteins, was believed to be responsible for the observed protection against challenge with a virulent strain of P. multocida. We have identified three lipoproteins and two additional proteins from a detergent-solubilized membrane preparation after trypsin digestion and peptide mass fingerprinting using MALDI-TOF mass spectrometry, lipoprotein B (PlpB, Mr of 30,233), lipoprotein (Mr of 37,374 kDa), and lipoprotein A (LipA, Mr of 79,738). The remaining two proteins identified included Putative Virulence factor (Mr of 117,437) and Porphyrin Biosynthetic Protein (HemY, Mr of 47,773). PlpB identity was confirmed by N-terminal sequence determination of C18-HPLC-purified peptides. Based on reactivity with a monoclonal antibody, the results indicate that the cross-protective antigen of P. multocida is Pasteurella lipoprotein B. |
