Interpretive Summary: Replication of Bluetongue (BTV) and epizootic hemorrhagic disease (EHDV) viruses in the primary target cells from cattle, sheep, black-tailed (BTD) and white-tailed (WTD) deer was compared. Purified cultures from the pulmonary arteries of cattle, sheep, BTD and WTD were used to determine if inherent species-specific are responsible for differences in expression of disease in BTV and EHDV-infected cattle, sheep, BTD and WTD. There were only minor differences in the replication, incidence of infection in BTV-17, EHDV-1 or EHDV-2 infected cattle, sheep, BTD and WTD cells. Differences in the expression of disease in BTV- and EHDV-infected ruminants are unlikely to reflect only species-specific differences in the susceptibility of their pulmonary artery cells to infection with these two orbiviruses.

Technical Abstract: Replication of Bluetongue (BTV) and epizootic hemorrhagic disease (EHDV) viruses in pulmonary artery endothelial cells (ECs) cultured from cattle, sheep, black-tailed (BTD) and white-tailed (WTD) deer was compared Purified EC cultures from the pulmonary arteries of cattle, sheep, BTD and WTD were used to determine if inherent species-specific differences in the susceptibility of ECs are responsible for differences in expression of disease in BTV and EHDV-infected cattle, sheep, BTD and WTD. The replication, incidence of infection and cytopathogenicity of prototype strains of BTV-17, EHDV-1 or EHDV-2 were compared in replicate cultures of ECs from each of the four ruminant species using virus titration and flow cytometric analysis. All three viruses replicated in ECs from cattle, sheep, BTD and WTD, however BTV-17 replicated more rapidly in ECs than did either serotype of EHDV. Each virus replicated to high titer in all EC cultures, although titers of EHDV-1 were significantly lower (P =0.018) in ovine ECs than the other cell types. Furthermore, all viruses caused extensive cytopathogenicity and a high incidence of cellular infection, however, the incidence of both cellular infection and cytopathic effect were lower in EHDV-1 infected ovine (P = 0.001) and EHDV-2 infected BTD EC cultures (P = 0.017). There were only minor differences in the replication, incidence of infection and cytopathogenicity in BTV-17, EHDV-1 or EHDV-2 infected cattle, sheep, BTD and WTD ECs. Differences in the expression of disease in BTV- and EHDV-infected ruminants are unlikely to reflect only species-specific differences in the susceptibility of their ECs to infection with these two orbiviruses
