Genetic Resources and Crop Evolution.52:245-247 Interpretive Summary: Two DNA marker methods (AFLP and SSR) were utilized to determine how genetically broad the collection of cultivated cowpeas and closely related plants are. These DNA marker methods did identify accessions that are closely related and accessions that are not pure or represent mixed or contaminated lines. The AFLP methodology was useful also for separating different species of Vigna. Neither method would successfully group the accessions of different subspecies and therefore the methods could not be used to determine whether the subspecies designations are really the correct grouping method. Technical Abstract: AFLP and SSR markers were utilized to determine the genetic diversity among Vigna unguiculata subspecies. The three AFLP primer sets and the 10 SSR primer sets successfully identified very closely related accessions and a lack of homogeneity in some accessions. The AFLP methodology was successful for separating different species of Vigna. However, the intra-subspecies variation was as great as the interspecies variation with both marker methods. Therefore, these methods with these numbers of primers could not group the subspecies into separate clades. |
