Interpretive Summary:

Technical Abstract: In order to determine if sufficient genetic variation exists within a population to permit selection for a particular trait, variation at the phenotypic level must also be quantifiable. Because a robust innate immune response is a favorable trait, our objectives were to determine if variation exists at the level of bacterial killing in bovine monocyte-derived macrophages (BMDM) challenged with E. Coli O157:H7, and to calculate if a significant association exists between bactericidal activity and previously determined bovine NOS2A haplotypes. Blood was collected from 164 head of cattle in the MARC breeding or feedlot populations. Monocytes were purified for assay, and genomic DNA was purified from the residual leukocytes. NOS2A haplotypes were analyzed using MALDI-TOF mass spectrometry, and bacterial killing was determined using a colorimetric assay with regression analysis. Percent bacterial killing by BMDM from 141 animals had a mean of 54.4%, CI95% 50.7-58.2% and the results were normally distributed. All NOS2A haplotypes previously identified in popular U.S. commercial cattle breeds were present in our study population at similar frequencies. Detailed analyses of the association of NOS2A haplotypes with bactericidal activity in BMDM will be presented.
