Interpretive Summary:

Technical Abstract: Organophosphate (OP) resistance has developed in Mexican populations of the southern cattle tick, Boophilus microplus. Reintroduction of this tick and its associated pathogens into the United States would cause severe economic losses to the cattle industry. We have begun a comparative study of the proteome of OP-susceptible and -resistant strains of B. microplus. As an initial step, we have exposed larvae from an OP-susceptible strain, Munoz, and an OP-resistant strain, San Roman, to 0.03 and 0.4% coumaphos, respectively, which are sublethal doses, well below the LD50 for each strain. Additionally, untreated larvae from each strain have also been collected. Membrane proteins were extracted from each sample and analyzed by 2-dimensional IEF-PAGE to identify OP-inducible proteins and strain-specific protein expression.
