Interpretive Summary: Commercial bacterins for the control of fowl cholera are comprised of P. multocida serotypes A:1, A:3, and A:4, which are the serotypes that are most often associated with outbreaks in poultry flocks. A membrane-associated protein fraction (cross-protective factor, CPF) containing 11 major proteins was reported to be responsible for the observed protection against challenge with a virulent strain of P. multocida. A 39-kDa protein was believed to be one of the proteins conferring protection. We have identified the 39-kDa protein as the lipoprotein B. This protein belongs to a family of bacterial lipoproteins that may be involved in bacterial virulence. Four additional proteins were identified, including a high molecular weight putative virulence factor, capsular biosynthetic protein, a porphyrin biosynthetic protein, and a protein involved in cell membrane biogenesis. By using peptide mass fingerprinting and MALDI-TOF mass spectrometry, we have identified two lipoproteins (lipoprotein B, PlpB, Mr of 30,233 and an unknown lipoprotein, Mr of 37,374 kDa) and three membrane-associated proteins (lipase A, LipA/PhyA, Mr of 79,738; putative virulence factor, Mr of 117,437; and porphyrin biosynthetic protein, HemY, Mr of 47,773). Our data suggested that the 39-kDa protein was identical to PlpB. Protein identity was confirmed by N-terminal sequence determination of two HPLC-purified tryptic peptides. PlpB belongs to the pfam03180 family of bacterial lipoproteins that may be involved with bacterial virulence. |
