Interpretive Summary: Surplus vegetable oils represent attractive renewable resources for the production of high-value food and industrial products. We are investigating microbial and enzymatic systems to utilize the large annual surplus of vegetable oils. Previously, we investigated lipase, an enzyme capable of modifying oil structure, from a variety of microbial sources for new applications. A collaborative study subsequently was carried out in the Reaction and Separation Laboratory of National Food Research Institute, Tsukuba City, Japan, to improve the activity of lipase toward vegetable oil, as supported by a short-term visit grant from the Scientific Cooperation Program, Foreign Agricultural Service, U.S. Department of Agriculture. It was found that microbial lipase activity could be improved by a unique modification method. Further characterizations of enzyme activity and molecular structure were conducted to provide new insights and techniques to researchers for developing industrial applications in the production of valuable structured lipids from vegetable oils.

Technical Abstract: Chromobacterium viscosum lipase was modified by stearic acid in two-phase system for improved trans-easterification (TE) activity. The stearic acid, dissolved in n-hexane, was mixed with lipase in buffer phase for 24 h. The modified lipase was present as insoluble layer at the interface, showed 9 units trans-esterification activity. The FTIR spectrum of lipase showed a new sharp peak corresponding to 1700 cm-1 as a result of stearic acid modification. The modified lipase catalyzed trans-esterification is used in preparing structured lipid, with capric rich oleic triglyceride as a model system.
