Interpretive Summary: Technical Abstract: In order to exploit full potential of economically available lignocellulosic biomass, Escherichia coli FBR5, a bacterial strain capable of producing ethanol efficiently in continuous cultures, was studied for substrate and product inhibition. The inhibition studies were performed under batch conditions. The culture can tolerate a maximum xylose concentration of 200 gL**-1; however, with a poor cell growth. The substrate inhibition studies were performed to observe cell growth and ethanol production. Similarly, ethanol inhibition studies were also performed. At an ethanol concentration of 60 gL**-1, no cell growth was observed. Further studies were extended to study the effect of medium component(s) on cell growth and ethanol production. Kinetic parameters (Saturation constant, Ks and Michaelis Menten constant, Km) that are essential for process scale up were measured. These studies were performed in order to be able to integrate fermentation with product recovery. Such a process integration benefits process economics. |
