Interpretive Summary:

Technical Abstract: Two screening assays were developed for the detection of tetracyclines in chicken muscle. The first method relies on monitoring the simple fluorescence of crude sample extracts in basic pH in the presence of a divalent cation. This rapid approach allows for efficient screening at the U.S. tolerance level (2 mg/kg). The second method uses time resolved fluorescence. While this method requires some sample clean up via solid phase extraction, it provides much higher sensitivity, allowing for screening at levels of 100 ng/g or below. Blind samples containing either tetracycline, oxytetracycline, or chlortetracycline were prepared and analyzed by the simple fluorescence assay for false positive and false negative results. Similarly, blind samples containing chlortetracycline were analyzed by the time resolved fluorescence method. Both the simple fluorescence and the time resolved fluorescence methods are able to provide useful screening, with the specific benefits of speed and high sensitivity, respectively.
