Interpretive Summary:

Technical Abstract: Marker-assisted selection (MAS) and analysis of quantitative trait loci (QTL) are becoming more common in plant breeding. These techniques are often limited by the random nature of many molecular markers, such as restriction fragment length polymorphisms (RFLP) and simple sequence repeats (SSR). Molecular markers for candidate genes would permit testing the linkage of those genes to desired traits. Sugarcane genotypes are polyploid, interspecific hybrids of the genus Saccharum. In sugarcane, one of the genes for sucrose synthase, Sus2, is hypothesized to be linked to sucrose concentration in stalks. A comparison of Sus2 gene sequences among diverse genotypes revealed insertion-deletion (indel) polymorphisms in the promoter region of the gene, and a polymorphic SSR in intron 14. These polymorphisms were used to develop PCR markers. Particular indel and SSR markers were specific to different species in the genus Saccharum. We are using the markers to test the linkage between Sus2 and sucrose concentration. If marker linkage to sucrose concentration is proved, these markers will allow selection for desirable Sus2 alleles and thus high sucrose concentration.
