Interpretive Summary:

Technical Abstract: An antibody was developed for use in an ELISA for the beta-agonist zilpaterol, a growth promoter/leanness enhancer approved in some countries but banned in others. The ELISA had optimal sensitivity at pH 7.4 with 0.3M salt concentrations with an IC50 of 3.94 ± 0.48 ng/mL. The assay demonstrated specificity for zilpaterol with no cross reactivity occurring with bamethane, clenbuterol, isoproterenol, metaproterenol, salbutamol, dobutamine, fenoterol, isoxsuprine, ractopamine, or salmeterol. The assay showed tolerance for up to 15% acetonitrile or DMSO and up to 10% acetone, methanol, and ethanol. Porcine and bovine urine caused qualitatively similar effects on the assay, although bovine urine resulted in greater decreases in the assay sensitivity as the percentage of urine in the assay was increased.
