Interpretive Summary: Expressed DNA sequences were taken from 47 cDNA libraries and were physically mapped, using genetic stocks and deletion lines in hexaploid wheat, to specific chromosome regions of wheat group 1 chromosomes. We found that the expressed sequences were not scattered from one end to the other of the group 1 chromosomes, but seemed to be clustered (gene-rich) in more distal regions of the chromosomes. The centromeric regions of all the chromosomes appeared to be gene-poor regions. This information will be of value to wheat geneticists utilizing expressed sequence locations in their research programs. Technical Abstract: A total of 944 expressed sequence tags (ESTs) and the corresponding 2,212 EST loci were mapped to homoeologous group 1 chromosomes in hexaploid wheat (Triticum aestivum L.). Expressed sequence tag deletion maps and the consensus deletion map of group 1 chromosomes were constructed to show EST distribution and EST density. Expressed sequence tag loci were unevenly distributed among chromosomes 1A, 1B, and 1D with 660, 826, and 726, respectively. The number of EST loci on the long arm outnumbered that for the short arm of all three chromosomes, with this arm difference basically attributed to the physical size of chromosome arms for 1A and 1B, and additional effects for 1D. The distribution of ESTs along chromosome arms was non-random with EST clusters occurring in the distal regions of short arms and middle regions of long arms, and low EST density in the proximal regions. Duplications of group 1 ESTs occurred at a high rate (35.49%) in other homoeologous groups. Seventy-five percent of wheat chromosome 1 ESTs had significant matches with rice sequences (E'e^-10), where large regions of conservation occurred between wheat consensus chromosome 1 and rice chromosome 5, and between the proximal portion of the long arm of wheat consensus chromosome 1 and rice chromosome 10. Only 9.5% of group 1 ESTs showed significant matches to Arabidopsis genome sequences. The results presented are useful for gene mapping, and evolutionary and comparative genomics of grasses. |
