Interpretive Summary:

Technical Abstract: Barley yellow dwarf is the most destructive viral disease in small grains worldwide. In 2002 and 2003, Alaskan Barley yellow dwarf virus (BYDV)-PAV and Cereal yellow dwarf virus (CYDV)-RPV were detected in barley (Hordeum vulgare) and oats (Avena sativa) by RT-PCR and ELISA. PCR products spanning the viral coat protein gene of 73 isolates were cloned and sequenced. Most (44/49) PAV isolates were similar to PAV-129 with the remainder related to PAV-NY. The RPV isolates clustered in three groups: 13 resembled an RPV isolate from Mexico, 5 were closely related to RPV-NY, and 6 formed a group that was distinct from any CYDV sequence currently in GenBank. The mean within-group sequence diversities were less than I percent for each of the 2 Alaskan PAV and 3 Alaskan RPV genotype clusters.
