Cross reaction of anti-human cd monoclonal antibodies on swine cells: a summary of the swine secton of the hlda8 animal homologues data. Meeting Abstract. Interpretive Summary: Technical Abstract: For the Animal Homologues section of the human leukocyte differentiation antigen (HLDA8) workshop our laboratories analyzed the reactivity on swine cells of the full panel of 386 commercially available mAb against human CD molecules from various companies. All mAb were screened for positive reactivity using flow cytometry on appropriate cell targets; ~80 of the mAb appeared to be positive on pig cells. Positive mAb were analyzed in more depth by confirming appropriate tissue and cell subset expression, effect of activation on expression, mw of antigen, and, if available, reaction with cloned swine gene product. To date, our results have confirmed that certain anti-human CD1b, CD9, CD14, CD18, CD29, CD41, CD44, CD45, CD47, CD49b,c, CD59, CD61, CD68, CD80, CD86, CD91, CD95, CD163, and CD172a mAb exhibit clear cross reactions with swine cells. Still other anti-CD mAb need to have their CD specificity confirmed. Some of these mAb will be unique tools for swine immune studies; others will add to the repertoire of commercial reagents available for swine. These cross-reactions will assist in defining an aim of this HLDA8 section, i.e., to identify mAbs which recognize conserved, species independent CD epitopes.
