Interpretive Summary: Technical Abstract: The microbial synthesis of xylitol from L-arabinose, which is one of the major components in hemicellulose, is useful for economical production of xylitol. It is known that fungi metabolize L-arabitol to D-xylulose 5-phosphate via xylitol. We are attempting to clone the genes from L-arabinose utilizing yeasts and to convert L-arabinose to xylitol by using recombinant Escherichia coli. L-xylulose reductase, which is one of three enzymes in the L-arabinose to xylitol pathway, has been cloned from Ambrosiozyma monospora and introduced in E. coli. As a result, this enzyme was functionally expressed in E. coli, and in vivo conversion of xylitol to xylulose (reverse reaction) was observed. |
