Interpretive Summary:

Technical Abstract: Gene expression profiles in resistant and susceptible soybean [Glycine max (L.) Merrill] in response to the soybean aphid, Aphis glycines Matsumura (Homoptera: Aphididae), were compared at 6 and 12 h after aphid infestation using cDNA microarrays representing over 18,000 soybean-expressed sequence tags. More genes were induced in resistant than susceptible soybean at 6 h after infestation. Genes that were differentially expressed between aphid and control treatments were selected as aphid-response genes. Eighty-four genes showed specific responses only in resistant soybean and included genes related to defense and a number of other activities. Expression patterns of three defense-related genes were examined at 6, 12, 24, 48, and 72 h after infestation in resistant and susceptible soybean by quantitative real-time PCR. The induction of the three defense-related genes in resistant versus susceptible soybean plants was earlier and stronger at 6, 12 and 24 h after infestation. Non-induced expression differences between the resistant and susceptible cultivars were also determined. Five genes with constitutively higher expression levels in the resistant cultivar were further analyzed for their potential roles associated with aphid resistance.
