Interpretive Summary:

Technical Abstract: Beet curly top virus (BCTV) is a persistent disease problem for bean production in the Pacific Northwest region and other dry areas where the virus and its vector, Circulifer tenellus are present. Breeding for resistance has been difficult because screening of selected lines in the field results in usually sporadic non-uniform infections across and within test plots. In order to circumvent the lengthy process required for identification of genetic resistance in bean, DNA markers have been developed that are tightly linked to the BCTV resistance gene(s) and have applications for marker-assisted selection (MAS). Separate DNAs bulked from eight curly top-resistant recombinant in-lines (RILS) and eight curly top-susceptible RILs were initially screened with 750 random decamer RAPD primers. A 1550 bp RAPD marker that selectively amplified products present in the bulk DNAs was converted to a SCAR marker to facilitate high throughput MAS of Bct-1 resistance gene across laboratories and breeding programs. Evaluation of the entire RIL population resulted in the marker present in 55 of 60 resistant and absent in 35 of 40 susceptible lines (90% co-segregation).
