Interpretive Summary: Technical Abstract: We have sequenced the genome of maize fine streak virus (MFSV), a member of the genus Nucleorhabdovirus in the family Rhabdoviridae. The genome has seven open reading frames (ORFs) on the viral complementary strand. Northern blot analysis showed that all seven genes are expressed in maize after inoculation with MFSV. Based on the sequence similarity with other rhabdoviruses, ORF1, ORF6, and ORF7 likely encode the nucleocapsid protein (N), glycoprotein (G), and polymerase (L), respectively. The sequences of ORF2, ORF3, ORF4 and ORF5 have no significant similarity with other rhabdoviruses. Using computer-assisted algorithms, a putative nuclear localization signal (NLS) was identified in the N gene and a transmembrane domain was identified in the G gene. ORF4 and ORF5 also contain putative NLS. Green fluorescent protein- fusions with ORF 2-5 were transiently expressed in tobacco leaves to determine which ORFs encode the nuclear-localized phosphoprotein (P) and matrix (M) proteins. Results show that ORF5 likely encodes M because it targets the nuclei of plant cells. Further, ORF2 apparently encodes P, because it is localized to the nuclei when coexpressed with N. The functions of ORF3 and ORF4 remain to be determined. These results suggest that the MFSV genome organization is 3'-N-P-3-4-M-G-L-5'. |
