Interpretive Summary:

Technical Abstract: The effect of the anesthetic methane tricainesulfonate (MS-222) (buffered with sodium bicarbonate and unbuffered) was evaluated on anesthetization efficacy and stress responses of juvenile channel catfish (18.4±2.1 g). Fish were exposed to MS-222 at 0, 90, 120, or 180 mg/L for 10 min after reaching stage 3 anesthetization. Blood was sampled immediately after exposure and after 30, 60, 120, 240, and 480 min recovery to measure blood glucose and plasma cortisol and osmolality. Time to reach stage 3 anesthetization was significantly longer and shorter for fish exposed to 90 mg/L and 180 mg/L MS-222, respectively, while recovery time was significantly shorter for 90 mg/L and longer for 180 mg/L. Buffering effectively neutralized acidity caused by MS-222 in anesthetization water but did not affect measures of stress. Concentration of MS-222 had a significant effect on the stress responses, but no consistent pattern was observed. The highest and lowest values were observed for MS-222 doses of 0 mg/L and 90 mg/L for cortisol, 180 mg/L and 0 mg/L for glucose, and 180 mg/L and 90 mg/L for osmolality. Because anesthetization is affected by water chemistry and fish size, further research is needed to determine optimal MS-222 dosages for channel catfish in aquaculture.
