Technical Abstract: We have described a protocol for the detection of two viruses of stone fruits, Cherry green ring mottle virus and Cherry necrotic rusty mottle viruses, both unassigned members of the Flexiviridae family of plant viruses. The protocol is useful for the detection of these viruses in budwood or other samples of stone fruits that may not be symptomatic. Our method is based on reverse transcription of the RNA genomes of the viruses followed by amplification with polymerase chain reaction (RT-PCR). The protocol can be completed in one or two days as compared to several months for the previously preferred method of graft indexing to susceptible cultivars of peach or cherry. We expect the method will be widely used by the research community and by others who are interested in the detection and/or characterization of this group of plant viruses.
