Interpretive Summary: Technical Abstract: The shotgun sequencing of the B. abortus vaccine strain, RB51 genome is nearly complete. Thus far, approximately 49,000 recombinant clones have been sequenced, generating approximately 34,300,000-bp of raw DNA sequence data. The resulting data has been compiled and aligned using the B. abortus strain 9-941 genome as a scaffold. A draft sequence of Chromosome I (approximately 2,122,270-bp) and Chromosome II (approximately 1,161,990-bp) has been generated. Greater than 85% of the sequence is high quality with multiple reads; the remaining sequence will need to be confirmed with additional analysis. A cursory comparison of the high quality portions of the RB51 genome sequence with that of B. abortus strain 9-941 shows greater than 99.78% sequence homology between the Chromosome I sequences, and 99.95% sequence homology between the Chromosome II sequences, excluding the highly variable VNTR loci and the presence of two additional copies of IS711 in the RB51 genome. The polymorphisms observed in RB51 Chromosome I are composed of 128 single nucleotide exchanges, and 4057 inserted/deleted nucleotides distributed across 29 loci. In Chromosome II, there are 67 single nucleotide changes, and 375 inserted/deleted nucleotides distributed across 12 loci. Key words: B. abortus; RB51; strain 9-941; genome |
