Interpretive Summary: Studies of the process whereby fat cells become non-fat cells capable of cell division are limited by the availability of appropriate approaches and methods. The methods and approach necessary to study this process are detailed and illustrated. The availability of this technology will allow critical examination of the process of fat cells converting to cells capable of cell division. Understanding regulation of this process will ultimately lead to means to control fat deposition in the meat animal.

Technical Abstract: In the present manuscript, the methods required to generate purified cultures of mature adipocytes, as well as stromal vascular cells, from the same isolation are detailed. Also, we describe the in vitro conditions for the dedifferentiation of the isolated mature adipocytes. These two types of cells may be used to reevaluate differences between presently available cellular models for lipogenesis/lipolysis and might provide a new cellular physiological system for studies utilizing the proliferative progeny from mature adipocyte dedifferentiation. Alternative possibilities to the dedifferentiation phenomenon are proposed, as this new area of research is novel.
