Interpretive Summary:

Technical Abstract: Acidovorax avenae subsp.citrulli (Aac), the causal agent of watermelon fruit blotch, was identified in watermelon in China in 2000 (T. Zhao et al., Acta Phytopathol. Sinica 4: 357-64. 2002). Although a considerable amount of watermelon and melon seed is produced in China, no information is available on the genetic diversity of strains of Aac in China. Thirty-two strains of Aac from China and one each representing PFGE haplotypes C, E, and G (sensu Wlcott et al., 2000. Phytopathology 90:191-96) were fingerprinted using AFLP. Bacteria were grown overnight in liquid shake culture and genomic DNA isolated by a modified Marmur method and digested with a combination of EcoRI/MseI restriction enzymes. The corresponding adaptors were ligated to the restricted DNA, and selective amplification was performed with Mse + C and EcoRI+0 primers. Preliminary results using GelCompar III software and 80% or greater relatedness assigned the 32 strains into 9 clusters. AFLP clusters V and VI contained PFGE haplotypes G and E, and C, respectively. All except strain 460 were related at 65% or greater and most (60%) belonged to cluster I.
