Interpretive Summary:

Technical Abstract: Zucchini yellow mosaic virus (ZYMV)-resistance was previously identified in Citrullus lanatus var. lanatus U.S. Plant Introduction (PI) 595203, and inheritance studies indicated that the ZYMV-resistance is conferred by a single recessive gene. In this study we examined the inheritance of resistance to the original ZYMV Florida strain (ZYMV-FL) using F1, F2 and reciprocal BC1 populations derived from PI 595203 and the watermelon cultivar ‘New Hampshire Midget’(NHM). The results confirmed that PI 595203 is resistant to ZYMV-FL and the resistance was conferred by a single recessive gene (zym). A study was initiated to identify DNA markers closely linked to the gene conferring the ZYMV resistance. Genomic DNA was isolated from the parental (PI 595203 and NHM) as well as their F1, F2 and BC1 plants. Two-hundred and seventy sequence-related amplified polymorphism (SRAP) markers were tested for polymorphism between the parents (PI 595203 and NHM). Of these, 135 markers appeared to be polymorphic, and are being used (with the mapping BC1 and F2 populations) for constructing a genetic linkage map and locating the ZYMV-resistance gene locus.
