Interpretive Summary: Technical Abstract: Gas chromatographic (GC) methods based on ECD, FID or MS detection are the most widely used methods for quantitative determination of the more toxic type-A trichothecenes (T-2 and HT-2 toxins). Deoxynivalenol (DON), the main type-B trichothecene frequently occurring in cereal grains, is commonly detected by HPLC/UV at ppb levels with good accuracy and precision. Recently, we have developed a method for the simultaneous determination of T-2 and HT-2 toxins in cereal grains using immunoaffinity column clean-up, labeling with 1-anthroylnitrile, and HPLC with fluorescence detection (FD). Moreover, a fluorescence polarization immunoassay (FPIA) has been used for rapid quantification of DON in wheat. Ongoing research efforts focus on improvement of the HPLC/FD method for T-2 and HT-2 using new labeling reagents (i.e. 1-naphthoyl chloride, 2-naphthoyl chloride, pyrene-1-carbonyl cyanide), and the optimization of FPIA for rapid screening of DON in soft and durum wheat kernels, semolina and pasta. |
