Interpretive Summary: Technical Abstract: Citrus flavonoids have been investigated for their biological actions in animals, particularly for their anti-cancer effects, and for their cholesterol-lowering and anti-inflammatory properties. An investigation of the production of cytokines in bacterial lipopolysaccharide (LPS)-treated human monocytes, has shown that the lipophilic polymethoxylated flavones (PMFs) are potent inhibitors of the production of the strongly pro-inflammatory tumor necrosis factor-alpha (TNF-alpha). In contrast, no inhibition was observed with the polar flavanone and flavone glycosides. A recent investigation of TNF-alpha production in LPS-administered mice showed that the PMF, 3’,4’,3,5,6,7,8-heptamethoxyflavone (HMF) administered i.p. dose dependently decreased TNF-alpha in the blood sera of treated animals. Subsequent HPLC-ESI-MS analysis of the blood sera showed the presence of HMF, as well as glucuronidated monohydroxy-hexamethoxyflavones and dihydroxy-pentamethoxyflavones. |
