Technical Abstract: Tristetraprolin (TTP) family of CCCH tandem zinc finger proteins (Zfp36, Zfp36L2, Zfp36L3) binds to and destabilizes some AU-rich element-containing MRNA's. TTP is a highly inducible gene but less is known about the expression of the other three homologues. The present study analyzed the relative abudance of the TTP family mRNAs and the effect of insulin on their mRNA levels and related gene expression in mouse 3T3-L1 adipocytes and macrophage RAW264.7 cells. Quantitative real-time PCR assays showed that Zfp36L1 and Zfp36L2 mRNAs were the two major forms and were approximately 8-10 fold of TTP in the adipocytes, but Zfp36L2 mRNA was the major one and was 4 and 10 fold of TTP and Zfp36L1 in RAW cells. Zfp36L3 mRNA was detected less than 1% of TTP mRNA levels in the adipocytes and RAW cells. Insulin increased TTP mRNA levels in the adipocytes, but decreased those of the three TTP homologues after 30-min treatment. Insulin did not affect the TTP family mRNA levels in RAW cells. Immunoblotting showed that TTP protein was also induced by insulin. However, the increased TTP protein did not appear to have significant effects on the steady state levels of its putative destabilizing targets including TNF, GM-CSF, and COX2 mRNAs in the adipocytes. This study demonstrated that TTP mRNA and protein were induced by insulin in the adipocytes and that Zfp36L3 mRNA was detected for the first time in cells other than mouse placenta and extraembryonic tissues.
