Interpretive Summary:

Technical Abstract: Recent mutational strategies for generating and screening of genes for optimized traits, including directed evolution, domain shuffling, random mutagenesis, and site-directed mutagenesis, have been adapted for automated platforms. Here we discuss the amino acid scanning mutational strategy and its adaptation to the plasmid–based functional proteomic workcell for use in rapidly generating optimized genes with multiple nucleotide changes to engineer improved protein and peptide functional characteristics.
