Fine Mapping the Slick Hair Locus in Senepol Cattle Using Three Additional Microsatellite Markers [abstract].Annual International Plant & Animal Genome Conference.January 16, 2008, San Diego, California, Final Abstracts Guide,p253:paper No.539 Interpretive Summary: Technical Abstract: The slick hair coat phenotype has been observed in tropical breeds of Bos Taurus Cattle in the New World, and has been studied in Senepol and Carora cattle that possess very short, sleek hair coats. The slick hair phenotype is known to be beneficial for heat tolerance in these breeds with body temperatures often 0.5 degrees Celsius lower during hot summer days for slick haired animals compared to their normal haired half-sibs. The slick hair coat locus has been mapped recently to BTA 20 in Senepol-derived cattle with a maximum LOD score of 9.4 for a 4.4 cM support interval between microsatellite markers DIK2416 and BM4107. In order to fine map the region containing the slick hair coat locus, three new microsatellite markers were designed from genomic sequences in this region and further evaluated in a population of unrelated Senepol (n=68) and Romosinuano (n=6) cattle. Additionally, previous genotypes from four other microsatellite markers that define this region of interest were analyzed using PHASE, which uses a Baysean method of haplotype reconstruction in unrelated individuals. The newly defined haplotype block was used to narrow the confidence interval around the slick hair coat locus and was used to identify candidate genes from the newly released Btau 4.0 genome assembly. |
