Title: Soybean Aphid Resistance in PI 243540 is Controlled by a Single Dominant Gene

Interpretive Summary: The soybean aphid (Aphis glycines Matsumura) is a recent pest of soybean [Glycine max (L.) Merr.] in North America. All current soybean cultivars in the USA are susceptible to this new pest. Insect resistance of plants is a very useful component of an integrated pest management program to control an insect problem. A maturity group (MG) IV plant introduction (PI) 243540 showed strong resistance against the Ohio collections of the soybean aphid. The objective of this study was to determine the inheritance of soybean aphid resistance gene(s) in PI 243540. The F1, F2 and F2-derived F3 families from a cross between an aphid susceptible cultivar ‘Wyandot’ and resistant PI 243540 were screened in greenhouse choice tests with the Ohio collection of the aphid. All F1 plants were resistant to the aphid. Of the 346 F2 plants, 264 were resistant and 82 were susceptible thus tightly fitting to a 3 resistant to 1 susceptible ratio (P=0.58) for inheritance of a single dominant gene for aphid resistance in the population. Similarly, 331 F2:3 families from this population segregated into 92 homozygous resistant : 157 heterozygous resistant : 82 susceptible families to fit into the expected 1:2:1 ratio (P=0.48). These results confirmed that a single dominant gene controlled the aphid resistance in PI 243540. The simple inheritance of this gene should be helpful in developing aphid resistant soybean cultivars by quickly transferring the gene to susceptible commercial cultivars using the backcross breeding approach. The aphid resistance gene from PI 243540 will be very useful to breeders and scientists interested in aphid resistance of soybean.
