Interpretive Summary: To identify the location of the binding sites on a major peanut allergy by human antibodies, human immune cells were cultured. The antibodies they produced were harvested and analyzed for binding to Ara h 1, a major peanut allergen. A unique antibody binding area was identified between amino acid 171 and 230 of the allergen and was named “area c”. Technical Abstract: To analyze epitopes of peanut allergen Ara h1, Epstein-Barr virus-transformed human peripheral oligoclonal B-cells were cultured to obtain antibodies to Ara h1. The combined reaction pattern with six oligoclonal antibodies showed there were six antibody binding areas named a to f in Ara h1. We found the novel antibody binding area named ‘‘area c’’ (171–230aa). |
