Interpretive Summary: Technical Abstract: We used a two-way pseudo-testcross mapping strategy in combination with simple sequence repeat (SSR) and Amplified Fragment Length Polymorphism (AFLP) markers to construct a genetic map of Hydrangea macrophylla. Map was generated using JoinMap 4.0 software, with a minimum LOD of 3.0 and a maximum recombination fraction of 0.4. Mapping populations were developed from reciprocal crosses between two divergent cultivars, “Bailmer” and “Veitchii”. “Bailmer”, which is sold as Endless Summer, is a remontant plant with mophead type flowers and is susceptible to powdery mildew. “Veitchii” is resistant to powdery mildew, has lacecap flowers, and does not rebloom. Preliminary analysis using 40 SSR loci as anchor markers was presented at the International Hydrangea Conference but had limited resolution. AFLP markers were recently added for greater marker saturation. |
