Interpretive Summary: This paper presents the sequence information on two new ilarviruses that infect raspberry, blackberry and strawberry crops. The two viruses, Blackberry chlorotic ringspot (BCRV) and Strawberry necrotic shock virus (SNSV) are members of subgroup 1 of the ilarviruses. These viruses were compared with the other two members of subgroup 1, Tobacco streak virus and Parietaria mottle virus (PMoV) at the molecular level. In addition, RT-PCR tests were developed for the detection of the two new viruses. SNSV and TSV infect strawberry and RT-PCR tests that detect both viruses or each virus individually were developed for diagnostic purposes. Using the sequence information obtained, it was shown that Black raspberry latent virus reported in the 1960's is actually the same virus as SNSV. The information presented clarifies the confusion of multiple virus names in the literature. Hopefully, quarantine regulations will be altered to include these virus names, since Black raspberry latent virus does not exist. SNSV and TSV infect strawberries, SNSV and BCRV infect raspberry and blackberry. The diagnostic tests developed will be useful for plant quarantine and certification programs. Technical Abstract: Three members of subgroup 1 of the genus Ilarvirus: blackberry chlorotic ringspot (BCRV), strawberry necrotic shock (SNSV) and tobacco streak viruses (TSV), may infect species of Rubus and Fragaria. Both BCRV and SNSV were identified recently but cause symptoms similar to those previously attributed to infection by TSV. The similarity of symptoms induced by the three viruses, and the degree of similarity observed between the sequence of TSV and the partial sequences of BCRV and SNSV, led us investigate the relationship between the three viruses and the other member of ilarvirus subgroup 1: Parietaria mottle virus (PMoV), at the molecular level. Here we present the complete genomic sequences of BCRV and SNSV and examine the phylogeny of the viruses included in subgroup 1 of the genus Ilarvirus identifying features at the molecular level that are either unique to subgroup 1 viruses or shared by other ilarviruses. |
