Interpretive Summary: Technical Abstract: Recombinant protein production is an important technology for antibody production, biochemical activity study, and structural determination during the post-genomic era. Limiting factors in recombinant protein production include low-level protein expression, protein precipitation, and loss of protein activity during the purification processes. This paper discusses several approaches to improve recombinant protein production, including 1) selecting high-level protein expression systems, 2) optimizing protein expression conditions, 3) minimizing protein purification steps, and 4) maintaining protein structures and functions. Finally, an example is presented for the expression and purification of anti-inflammatory protein tristetraprolin from E. coli and human cells. |
