Title: Synergistic inhibition of interleukin-6 production in adipose stem cells by tart cherry anthocyanins and atorvastatin

Interpretive Summary: In vitro data from our current study demonstrate that nutrient called flavonoid that are contained in a high levels in dark color fruit such as tart cherry as well as cholesterol lowering drug atorvastatin calcium (lipitor) have independent anti-inflammatory effects. To test whether adding together falvonoids and lipitor have potential additive benefits on lowering interleulin-6 (IL-6), a activator of well known cardiovascular risk factor c-reactive protein (CRP), we added tart cherry extracts and isolated individual flavnoids from tart cherry to adipose stems cells. Data from out study showed that LPS-induced adipose stem cell secretion of IL-6 is reduced with the addition of tart cherry extract, a mixture of tart cherry anthocyanins, and pure tart cherry cyanidin-3-O-glucoside (C3G) in a dose-dependent manner. Furthermore, lipitor and C3G exhibited additive effects in reducing LPS-induced IL-6 secretion from adipose stem cells. It is possible that combined this therapy may reduce necessary dosage and number of drugs used by patients undergoing treatment for CVD and likely decrease the number of adverse effects due to drug reactions.

Technical Abstract: Studies have shown positive correlations between inflammatory cytokines such as interleukin-6 (IL-6) and the development of chronic diseases including cardiovascular disease by activating C-reactive prorein (CRP). Both atorvastatin calcium (lipitor) as well as flavonoid rich fruit such as tart cherry has potent anti-inflammatory effects on IL-6 secretion when added together with lipitor. In this study, we investigated whether tart cherry extract or any specific anthocyanins contained in the tart cherry have additive anti-inflammatory effects when added together with lipitor. Results showed that LPS-induced adipose stem cell secretion of IL-6 is reduced with the addition of tart cherry extract, a mixture of tart cherry anthocyanins, and pure tart cherry cyanidin-3-O-glucoside (C3G) in a dose-dependent manner. Furthermore, lipitor and C3G exhibited additive effects in reducing LPS-induced IL-6 secretion from adipose stem cells. In conclusion, these results support potential benefits of using dietary phytochemicals in conjunction with pharmacological therapies to decrease adipose inflammation, drug doses, and ultimately, drug-induced adverse effects.
