Interpretive Summary: This manuscript summarizes the results of part of the second round of analyses performed by scientists participating in the First International Swine CD Workshop. This workshop was set up to compare and standardize the reactivities of monoclonal antibodies (mAb) that recognize pig leukocyte cell surface markers. For the second round analyses, the reactivities of 54 mAbs on T cell subsets or lines were analyzed by flow cytometry and the data subjected to statistical clustering to identify group s of mAbs that reacted similarly with specific pig T cells. These studies identified 6 mAb as reactive with porcine CD5 based on the results of one- and two-color FCM analyses. These clustered CD5 mAbs were examined in more detail with the result t hat it was determined that they bound to the same area, or epitope, of the CD antigen. Because of lack of immunoprecipitation studies for five out of the six anti-CD5 mAb, to date only one mAb b53b7 can be definitely described as an anti-porcine CD5 mAb. The five other mAb included in this cluster are characterized by the same labeling pattern in FCM and the fact that they recognize the same or closely related epitopes, which has been demonstrated by blocking of biotinylated mAb b53b7. Without specific molecular mass of the antigen these mAb were allocated to the wCD5a subcluster.

Technical Abstract: Based on the results of one- and two-color FCM analyses and epitope blocking, six mAb specific for the porcine CD5 have been identified. Because of lack of immunoprecipitation studies for five out of the six mAb, to date only one mAb b53b7 (#119) can be definitely described as an anti-porcine CD5 mAb. The five other mAb included in this cluster are characterized by the same labeling pattern in FCM and the fact that they recognize the same or closely related epitopes, which has been demonstrated by blocking of biotinylated mAb b53b7. Without specific molecular mass of the antigen these mAb were allocated to the wCD5 subcluster.
