This workshop was set up to compare and standardize the reactivities of monoclonal antibodies (mAb) that recognize pig leukocyte cell surface markers to assign them specific cluster of differentiation numbers (CD#). For the second round analyses, the reactivities of 54 mAbs on T cell subsets or lines were analyzed by flow cytometry a nd the data subjected to statistical clustering to identify groups of mAbs that reacted similarly with specific pig T cells. These results confirmed the assignment of 4 monoclonal antibodies (mAb) to the CD4 T cell cluster. Clustered mAbs were examined in more detail to determine whether they bound to the same area, or epitope, of the CD antigen. Epitope analyses and the molecular weight of the precipitated antigen confirmed that 3 mAb (74-12-4, ,10.2H2, b38c6) bound to the same epitope, CD4a, of the swine antigen. One other mAb (PT90) appears to bind the same epitope but its antigen molecular weight has not been confirmed; it is noted as reacting with the wCD4a antigen. These results will enable researchers to readily compare results of their studies using porcine CD4 specific mAb and thus enrich swine immunologic studies. Technical Abstract: The initial results of the First International Swine CD Workshop confirmed the assignment of 4 monoclonal antibodies (mAb) to the CD4 T cell cluster.
