Interpretive Summary: This manuscript summarizes the results of part of the second round of analyses performed by scientists participating in the First International Swine CD Workshop. This workshop was set up to compare and standardize the reactivities of monoclonal antibodies (mAb) that recognize pig leukocyte cell surface markers to assign them specific cluster of differentiation numbers (CD#). This manuscript is an overview of the T-cell section, of the analyzed target cells, the participating laboratories and their respective tasks during the second round of the CD workshop. For the second round analyses, nine different laboratories analyzed the reactivities of 54 mAbs on T cell subsets or lines by flow cytometry and the data was then subjected to statistical clustering to identify groups of mAbs that reacted similarly with specific pig T cells. These cluster analyses revealed that 36 out of the 54 mAb could be distributed to 6 CD and 2 swine workshop clusters (SWC). Clustered mAbs were examined in more detail to determine whether they bound to the same area, or epitope, of the CD antigen in the later stages of the second round of workshop analyses. The following porcine clusters could be defined: CD2 (recognized by 5 mAb), CD4 (4 mAb), CD5 (6 mAb), CD6 (2 mAb), CD8 (6 mAb, recognizing 2 epitopes), CD25 (1 mAb), SWC1 (9 mAb) and SWC2 (2 mAb). These assignments will enable researchers to compare their scientific data using anti-pig T cell mAb much more readily.

Technical Abstract: The first round analyses of the first porcine CD workshop led to 54 mAb which were assigned, because of possible reactivity with T-lymphocyte specific antigens and/or activation antigens, to the T-cell section for further analyses. This manuscript is an overview of the T-cell section, the analyzed target cells, the participating laboratories and their respective tasks during the second round of the CD workshop. 54 mAb were tested in FCM for their reactivity on various porcine target cells by nine different laboratories. Cluster analyses performed with the Database IV program revealed that 36 out of the 54 mAb could be distributed to 6 CD and 2 preliminary workshop clusters. The following porcine clusters could be defined: CD2 (recognized by 5 mAb), CD4 (4 mAb), CD5 (6 mAb), CD6 (2 mAb), CD8 (6 mAb, recognizing 2 epitopes), CD25 (1 mAb), SWC1 (9 mAb) and SWC2 (2 mAb).
