Title: Identification of I-7 expands the repertoire of genes for resistance to Fusarium wilt in tomato to three resistance gene classes

Interpretive Summary: The Tomato gene I-7, that provides resistance to Fusarium wilt in tomato, was identified and shown to encode a signaling molecule. The identification of I-7 provides molecular markers for marker-assisted breeding for resistance to Fol race 3. The distinct Fol resistance characteristics conferred by I-7 make it a useful additional tool for the tomato breeder’s toolbox.

Technical Abstract: The tomato I-3 and I-7 genes confer resistance to Fusarium oxysporum f. sp. lycopersici (Fol) race 3 and both genes were introgressed into the cultivated tomato, Solanum lycopersicum, from the wild relative Solanum pennellii. I-3 was identified previously and encodes a S-receptor-like kinase, but little was known about I-7 and markers for breeding I-7 were lacking. We used RNA-seq and SNP-analysis to isolate I-7 and showed that it encodes a leucine-rich-repeat receptor-like protein (LRR-RLP) and like other RLPs is EDS1 dependent. We found that I-7 also confers resistance to Fol races 1 and 2. The identification of I-7 has allowed us to develop molecular markers for marker-assisted breeding of both I-3 and I-7 genes for resistance to Fol race 3. The distinct Fol resistance characteristics conferred by I-7 make it a useful additional tool for the tomato breeder’s toolbox.
