Thus, to facilitate genome sequencing, a selection technique known as Cot selection was used to isolate a less complex fraction of the genome for sequencing using 454 GS FLX technology. These sequences have been deposited at NCBI under the BioProject, BioSample, and Short Read Archive (SRA) accession numbers PRJNA412317, SAMN07988077, and SRS2669643, respectively. Technical Abstract: A reassociation kinetics-based approach was used to reduce the complexity of genomic DNA from the Deutsch laboratory strain of the cattle tick, Rhipicephalus microplus, to facilitate genome sequencing. Selected genomic DNA (Cot value = 660) was sequenced using 454 GS FLX technology, resulting in 356.1 Mbp of sequence data after quality filtering.
