Interpretive Summary: Chinese aconite (Aconitum carmichaelii) is species of the family Ranunculaceae native to East Asia. Aconites have been cultivated as an ornamental and medicinal plant for centuries, although the latter use requires great care because the plant contains highly poisonous alkaloid compounds, especially in the tubers. The crop is clonally propagated mainly by tubers, so its production is vulnerable to many diseases including those caused by viruses. In this study, a new virus was identified by high-throughput sequencing in association with diseased aconite plants in Yunnan, China. The sequence of the new virus, named Aconitum amalgavirus 1 (AcoAV-1), was determined, which confirmed it as a new member of the amalgavirus group. The virus was detected in 37 out of 88 symptomatic plants that were tested, indicating it may frequently infect aconite production fields. The genomic sequence and protocols developed in this study can be used to detect the virus, and help investigate whether it causes economic losses in aconite production.

Technical Abstract: A novel virus named Aconitum amalgavirus 1 (AcoAV-1) was identified from Chinese aconite (Aconitum carmichaelii) plants with disease symptoms. The genome of this monopartite dsRNA virus is 3,370 nucleotides long and contains two partially overlapping open reading frames (ORFs) that encode a putative coat protein and RNA dependent RNA polymerase (RdRp), respectively. AcoAV-1 shares amino acid sequence identities of 34.9-50.7% to other amalgaviruses at RdRp. Phylogenetic analyses group the new virus with blueberry latent virus and related viruses in a cluster, suggesting it is a member of the genus Amalgavirus in the family Amalgaviridae.
