Interpretive Summary:

Technical Abstract: Segregation and inheritance of two transgenes, uidA (B-glucuronidase) and bar, were studied in 25 families of genetically engineered oat. Molecular transgene integration patterns and expression of the transgene phenotypes were followed in two generations of progeny. Disomic Mendelian inheritance of both transgene phenotypes and the transgene integration patterns was observed in 20 families. The transgene segregation pattern of four of these families indicated the presence of two or more unlinked functional transgene loci. Five other families exhibited abnormal inheritance and segregation of the transgene phenotypes. In these abnormal families, the number of transgene-positive plants was significantly lower than expected and no transgene-positive homozygotes were recovered. Transgene silencing was observed in some families and may result from the multiple linked transgene integration sites and extensive transgene rearrangements detected din many of the transgenic families. The 80% frequency of transgenic families exhibiting Mendelian inheritance indicated that the transformation system will be useful in oat improvement.
